What it is

Semaglutide is a GLP-1 receptor agonist — a modified version of a gut hormone that regulates appetite and blood sugar. It's the active molecule in Ozempic (diabetes), Wegovy (weight), and Rybelsus (oral), which makes it the single most recognized peptide on earth. It has years of approved real-world use and enormous trial datasets — in research-chemical form it is the same molecule outside the pharmaceutical supply chain, which is why third-party testing matters doubly here.

The pharmaceutical titration is the template: 0.25 mg once weekly for 4 weeks, then 0.5 → 1.0 → 1.7 → 2.4 mg, stepping up no faster than monthly and holding at the lowest effective dose. Slower is smoother — nearly all the GI misery in this class comes from rushing the ladder. Protein and resistance training are the standard companions to protect muscle during loss.

Compared to its siblings: tirzepatide trials showed larger average loss, and retatrutide larger still (in phase 2) — semaglutide's edge is the longest approved track record and the gentlest introduction to the class.
